Particle size, dust content and attrition, friability testing

Particle size, dust content and attrition and friability relate to the risks of operator inhalation and equipment blockage when using products formulated as powders, granules or tablets. Attrition and friability (hardness and integrity) are determined using CIPAC MT 187 (powders), MT 178 (granules) and MT 193 (tablets); the amount of unbroken product is assessed after physical handling, for example in MT 178 the product is rolled in a glass bottle containing glass beads, then sieving and weighing the amount that has not broken up. If attrition is below a certain level, dustiness is measured by collecting and measuring airborne dust from a chamber in which granules have fallen (CIPAC MT 171), and if dust is >1% (w/w) then the dust size is also determined. 

Particle size distribution is determined by CIPAC MT 187 or OECD 110 for powders (using laser diffraction and microscopy, respectively), CIPAC MT 59.2 or 187 for granules and CIPAC MT 170 for water-dispersible granules. MT 59.2 and 170 determine particle size uses sieves of various sizes; inhalation toxicity testing may be needed if >1% is found to be <50 µm.
