The Covance Biomarker Center of Excellence is pleased to announce the immediate availability of three new technology solutions to better support high-sensitivity biomarker analysis for single studies or as part of a comprehensive discovery and translational services program. After obtaining high-sensitivity data using any of these three platforms, sponsors can make better informed decisions with the best chance of success to advance their therapeutic programs.
Gallios Flow Cytometer
The Gallios Flow Cytometer combines extraordinary sensitivity, resolution and dynamic range together with high-speed data collection. The Biomarker Center of Excellence's new Gallios is outfitted with three lasers (violet, blue, red) and configured to detect up to 10 fluorescence signals simultaneously. The instrument also includes an enhanced forward scatter detector for the most accurate size measurement, e.g. for microparticle studies. The Gallios may be used for analysis of samples in standard test tubes or can be connected to the HyperCyt sampling accessory from IntelliCyt Corp., for sampling from microwell plates.
HyperCyt sample handling technology transfers samples from the wells of a 96- or 384-well microplate to the flow cytometer in a rapid sequential fashion. This method is the most cost-effective, rapid and efficient way to screen compound or antibody libraries using flow cytometry. Coupled to the 10 color Gallios, the HyperCyt platform increases capacity for complex immunophenotyping panels and assays of immune function.
Erenna Immunoassay System
The Singulex Erenna immunoassay system is a magnetic microparticle-based immunoassay technology that employs microfluidic instrumentation capable of counting single molecules of a reporter fluorophore in solution. Background signal is reduced by eluting the analyte and detection antibody away from the magnetic microparticle solid support prior to analysis, hence enabling the detection of single molecule of analyte-detection antibody complex. The sensitivity gained by the use of this technology allows for the measurement of some endogenous biomarker levels difficult to detect by conventional immunochemical assays. A large dynamic range of 4+ logs is obtained by combining single-molecule counting (low range) with photon counting (mid range) and total light measurements (high range).
Hybrid Quadrupole Time-of-flight Tandem Mass Spectrometer
The LC/MS-based bioanalytical assays have been used on a routine basis for quantitation of xenobiotics, metabolite(s), endogenous analyte(s), biomarkers, etc. throughout the drug discovery and development process. Some of their merit features include great specificity and sensitivity, rapid turnaround, flexibility and cost efficiency for new assay development. By adhering to 'fit-for-purpose' in method development and validation for biomarker identification and quantitation, we provide an unique LC/MSMS analytical capability in supports of biomarker analysis for both preclinical and clinical drug development studies. The Biomarker Center of Excellence has a new hybrid quadrupole time-of-flight tandem mass spectrometer (AB SCIEX TripleTOF™ 5600 System) installed and ready for assay development and sample analysis. This instrument uniquely integrates comprehensive qualitative exploration, rapid profiling, and high-resolution quantitation workflows on a single platform. It combines the highest sensitivity detection (equivalent to high-performance triple quadrupole), high-resolution with at least 5x better acquisition speed, and stable mass accuracy over days of acquisition. The instrument provides a wide range of applications for both small molecules and large molecules, such as metabolites/protein identification and targeted quantification.
