In vitro production of monoclonal antibodies has increasingly become a more viable option for researchers. With more than 15 years of tissue culture experience, Covance Immunology Services has successfully employed numerous in vitro production methods. Our goal is simple: Match the characteristics of your cell line with the optimum production method, at an affordable cost. Covance has the experience and knowledge to achieve this goal, for both small and large-scale production runs, with the assurance of quality standards required by the research community. Many investigators choose to follow up with Protein A or Protein G purification to isolate the immunoglobulins and concentrate the antibody. Any historically successful media recipe will provide for optimal cell growth. If the recipe is not known, Covance staff must determine optimal cell growth conditions. This may lengthen production time.
We offer scale-up in vitro antibody production in roller bottles or small bioreactors as the final phase of a custom monoclonal project. As an alternative, you may provide an established hybridoma cell line for a production run.
We offer the following scale-up technologies:
- Roller Bottles — Scale up of your antibody through roller bottle production eliminates the possibility of interfering murine proteins as can be found with in vivo methods. The result is a cleaner antibody preparation.
- Hollow fiber — Covance, an industry leader, offers Hollow Fiber Technology for production of milligram to gram quantities of monoclonal antibodies. Our vast experience enables us to optimize your monoclonal cell line for maximum yields in cell growth and secreted protein production.
Although hollow fiber is an established technology, significant advances in fiber materials have greatly improved productivity allowing for denser cell growth. Benefits of hollow fiber production include the ability to produce high concentration quantities of mAbs, elimination of murine contaminants as well as the ability to adapt cells to serum free-media.
- Integra CL 350 or CL 1000 — These small bioreactors are a membrane based cultivation system intended for scale up of your antibody at a higher concentration than can be achieved through roller bottle production.
- Cell Factory — The factories are designed for large scale cell culture production. They are a good alternative for cells that like static environments and provide an increased surface area which ranges from 632 cm2 to 25,284 cm2 with volumes ranging from 200 milliliters to 8 liters.
